ABSTRACT

Felid alphaherpesvirus 1 (FeHV-1) and feline calicivirus (FCV) affect cats worldwide. The aim of this study was to evaluate the frequency of occurrence of FeHV-1 and FCV in cats with clinical signs of respiratory, oral and/or ocular disease. Samples were collected from cats cared for in veterinary ambulatory and clinics and submitted to molecular detection and viral isolation. Of the 49 cats evaluated, 45 (92%) were positive for at least one of the viruses; 82% (40/49) were positive for FeHV-1 and 41% (20/49) for FCV. Of these, 31% (15/49) were coinfection cases. For FeHV-1, 45% (18/40) of the cats tested were positive from the collection of eye swab, and the same percentage (9/20) was obtained for the FCV by the oral swab. FeHV-1 and/or FCV were isolated in 35% (17/49) of the samples. The main clinical sign observed was ocular secretion in 71% (35/49) of cats, characterized as mild serous, purulent or serosanguineous, and in some cases associated with ocular injury and marked chemosis. Our findings demonstrate the high occurrence of FeHV-1 and FCV in domestic cats in southern Brazil and indicate that measures should be implemented to improve the diagnostic, prevention and management against of these important diseases.(AU)


Alphaherpesvírus felídeo 1 (FeHV-1) e calicivírus felino (FCV) afetam gatos mundialmente. O objetivo deste estudo foi identificar a frequência de ocorrência de FeHV-1 e FCV em gatos com sinais clínicos de doença respiratória, oral e/ou ocular. Amostras foram coletadas de gatos atendidos em ambulatório e clínicas veterinárias e submetidas à detecção molecular e isolamento viral. Dos 49 gatos avaliados, 45 (92%) foram positivos para ao menos um dos vírus; 82% (40/49) foram positivos para o FeHV-1 e 41% (20/49) para o FCV. Destes, 31% (15/49) foram casos de coinfecção. Para o FeHV-1, 45% (18/40) dos gatos foram positivos na coleta do swab ocular, e o mesmo percentual (9/20) foi obtido para o FCV a partir do swab oral. FeHV-1 e/ou FCV foram isolados em 35% (17/49) das amostras. O principal sinal clínico observado foi secreção ocular em 71% (35/49) dos gatos, caracterizada como serosa, purulenta ou serossanguinolenta e, em alguns casos, associada à lesão e quemose. Nossos resultados demonstram a alta ocorrência de FeHV-1 e FCV em gatos domésticos na região Sul do Brasil e indicam que devem ser implementadas medidas para melhorar o diagnóstico, a prevenção e o manejo contra essas importantes doenças.(AU)

ABSTRACT

Bovine alphaherpesvirus type 1 (BoHV-1) is the etiological agent responsible for serious infections that compromise the respiratory and genital tracts of affected cattle. In order to estimate the seroprevalence of BoHV-1 and to identify the associated risk factors in dairy farms in the city of Senador Guiomard, Acre, Brazil, the present study was carried out through the analysis of 180 blood serum samples submitted to the screening of anti-BoHV-1 by the virus neutralization test (VN) and by means of the evaluation of the epidemiological questionnaire applied in the eight investigated properties. The prevalence of seropositivity for BoHV-1 was 61.1%, ranging from 43.3 to 86.2% among the analyzed properties. The variable absence of veterinary assistance showed statistically significant association (odds ratio ­ OR = 2.10; p < 0.001) with alphaherpesvirus infection. The results demonstrate that the frequency of BoHV-1 is high and needs to be controlled through prophylactic and health management measures.(AU)


O alfaherpesvírus bovino tipo 1 (BoHV-1) é o agente etiológico responsável por graves infecções que comprometem os tratos respiratório e genital dos bovinos acometidos. Com o objetivo de estimar a soroprevalência de BoHV-1 e identificar os fatores de risco associados em propriedades leiteiras do município de Senador Guiomard, Acre, foi realizado o presente estudo, por meio da análise de 180 amostras de soro sanguíneo submetidas à pesquisa de anticorpos anti-BoHV-1 pelo teste de vírus neutralização (VN) e por meio da avaliação do questionário epidemiológico aplicado nas oito propriedades investigadas. A prevalência de soropositividade para BoHV-1 foi de 61,1% variando de 43,3 a 86,2% entre as propriedades analisadas. A variável ausência de assistência veterinária apresentou associação estatisticamente significativa (odds ratio ­ OR = 2,10; p < 0,001) com a infecção pelo alfaherpesvírus. Os resultados demonstram que a frequência de BoHV-1 é alta e precisa ser controlada através de medidas profiláticas e de manejo sanitário.(AU)

ABSTRACT

After a long-term co-evolution, alphaherpesviruses have established mutual adaptability with their hosts. Some alphaherpesviruses have typical neurotropic characteristics, which have received extensive attention and in-depth research. Neurotropic alphaherpesviruses can break through the host barrier to infect neurons and multiply in large numbers in the neuron cell body to complete further proliferation or establish latent infection in the cell body. Either in the process of infecting neurons or further spreading, alphaherpesviruses will undergo transmission along axons or dendrites, so this process is an integral part of the life cycle of the viruses, and is also a key factor for the viruses to spread in nervous system. Therefore, studies on transportation of alphaherpesviruses in neurons will provide new insights of the viruses and promote the development of corresponding vaccines or targeted therapeutic pharmaceuticals. In addition, the neurotropism of alphaherpesviruses is conducive to the analysis of nerve circuits. Herein, the mechanisms of alphaherpesvirus transport in axons were reviewed, and the research direction and application of the transport of alphaherpesviruses in axons were put forward, which can provide reference for the prevention and control of alphaherpesviral infections.

ABSTRACT

To clarify the pathogenesis of Duck enteritis virus (DEV), the cDNA library of duck's liver infected by DEV and a bait plasmid containing DEV nucleocapsid protein (NP) gene were constructed, then the receptor was screened from the cDNA library plasmid by the yeast two-hybrid system and verified by GST pull-down test. The results showed that the capacity of the primary cDNA library was 1 x 106 CFU with insertion size from 0.5 to 1 kb, and the bait plasmid of pGBKT7-NP showed no self-activation. The receptor reacting with DEV NP in duck liver was initially confirmed as the protein kinase C inhibitor (PKCI). These results provide new clues for further investigation on pathogenesis of DEV.

ABSTRACT

Introduccion: Lippia alba (Mill) N.E. Brown es una planta aromatica perteneciente a la familia Verbenaceae, ampliamente usada en Suramerica y Norteamerica como infusion contra la hipertension, problemas digestivos, nauseas y resfriados. Objetivo: En el presente estudio, se evaluo la actividad antiviral in vitro frente al Herpesvirus Humano Tipo 1 (HSV-1), de veinte aceites esenciales de L. alba y, diez de sus componentes mayoritarios. Metodologia: La actividad antiviral in vitro fue evaluada empleando la t¨¦cnica modificada, de titulacion del punto final (EPTT). Los aceites esenciales, de plantas de L. alba recolectadas de diferentes regiones del pais, fueron obtenidos por hidrodestilacion asistida con radiacion de microondas (MWHD). Se determino por cromatografia de gases¨Cespectrometria de masas (GC-MS), la composicion quimica, de los veinte aceites esenciales de L. alba, identificandose dos quimiotipos: citral y carvona. Resultados y conclusiones: Los aceites quimiotipo carvona, BC1 y CA2, mostraron actividad antiherpetica in vitro, moderada sobre monocapa de celulas HeLa infectadas, con valores de Rf de 1x101,5, en concentraciones, de 250¦Ìg/mL y 125¦Ìg/mL, respectivamente. Los controles positivos, sulfato de heparina y aciclovir, redujeron el titulo viral con valores de Rf, en orden, de 1x102 y 1x104. Ninguno de los monoterpenos evaluados mostro actividad contra el HSV-1.


Introduction: Lippia alba (Mill.) N.E. Brown is an aromatic shrub belonging to the Verbenaceae family, which is widely used all over South and Central America as an infusion against, hypertension, digestive troubles, nausea and cold. The antiviral activity in vitro against Herpesvirus 1, Human (HSV-1) of twenty essential oils of L. alba and ten of its components were evaluated. Methodology: The antiviral activity was determined using a modified end-point titration technique (EPPT). The essential oils from L. alba collected in different regions of Colombia were obtained by microwave-assisted hydrodistillation (MWHD). Their chemical compositions were determined by GC and GC/MS. Two chemotypes were distinguished, characterized by carvone and citral as main constituents. Results and conclusions: Carvone chemotype oils BC1 and CA2 were found to be moderate antiviral activity over infected confluent monolayers of HeLa cells with Rf value of 1x101,5 at a concentration of 250¦Ìg/mL and 125¦Ìg/mL, respectively. Heparin sodium salt and acyclovir were used as positive controls and showed Rf values of 1x102 and 1x104, respectively. None of monoterpenes tested showed antiviral activity against HSV-1.

ABSTRACT

Aujeszky' s disease (AD) is an infectious disease causing important economic losses to the swine industry worldwide. The disease is caused by an alpha-herpesvirus, Aujeszky' s disease virus (ADV), an enveloped virus with a double stranded linear DNA genome. The ADV genome encodes 11 glycoproteins, which are major targets for the immune system of the host in response to the infection. The glycoprotein E (gE) is a non-essential protein and deletion of the gE gene has been used for the production of marker vaccines. Aiming to develop molecular reagents for the production of a gE specific ELISA test, the gE gene was amplified by PCR, cloned and expressed into a baculovirus expression system. The recombinant DNA vector pFastBac-gE-ADV was used for site-specific transposition into the recombinant baculovirus (bacmid). Colonies with recombinant bacmid-pFastBac-gE-ADV were selected by antibiotic and color selection and the presence of the gE gene was confirmed by PCR. The recombinant bacmid-pFastBac-gE-ADV was cotransfected in insect Trichoplusia ni and the presence of the recombinant DNA and gE protein were detected by PCR, SDS-PAGE and Western blotting, respectively.


A doença de Aujeszky (DA) é uma enfermidade infecto-contagiosa que causa grandes perdas econômicas ao produtor e à agroindústria suinícola em todo o mundo. É causada pelo vírus da doença de Aujeszky (VDA), um alfaherpesvírus envelopado com genoma DNA de fita dupla e linear. O genoma do VDA codifica 11 glicoproteínas, as quais são os maiores alvos do sistema imune do hospedeiro em resposta a infecção. A glicoproteína E (gE) é uma proteína não essencial e a deleção do gene da gE é muito utilizada para a produção de vacinas com marcadores. Com o objetivo de desenvolver insumos moleculares para a produção de um teste de ELISA específico para gE do VDA, a seqüência do gene da gE foi amplificada, clonada e expressa no sistema de expressão em baculovírus. O produto da amplificação foi clonado no vetor pGem®-T Easy e subclonado no plasmídeo de expressão pFastBac™1. O DNA recombinante pFastBac-gE-VDA foi usado para a transposição sítio-específica no baculovírus recombinante (bacmid). Após seleção por antibióticos e cor, as colônias com o recombinante bacmid-pFastBac-gE-VDA foram selecionadas e a presença do gene da gE foi confirmada por PCR. O DNA recombinante viral, bacmid-pFastBac-gE-VDA, foi usado para cotransfecção de células de inseto Trichoplusia ni e a presença do recombinante e a proteína gE foi determinada por PCR, por SDS-PAGE e Western blotting, respectivamente.

ABSTRACT

The first step of the herpes virus infection is the attachment to heparan sulfate molecules on the cellular membrane. In order to improve the characterization of this phenomenon, we compared the inhibitory effect of six sulfated polyelectrolytes (PE): heparin, heparan, low molecular wight heparin, chondroitin, dextran and protamine on plaque formation by pseudorabies virus (PRV) were compared. The PE with the highest antiherpetic effect was heparin, followed by dextran sulfate. Heparan sulfate, which has been proposed as the initial receptor of herpes virus on the cell surface showed and effect 100-fold lower than heparin. Comparative inhibition curves of heparin and heparan sulfate against three herpes viruses: herpes simplex virus 1 (HSV-1), HSV 2 and PRV showed similar kinetics of inhibition of plaque formation, suggesting these viruses could share similar cell adsorption mechanisms
